No information about Greene’s art training has yet been found. His name change from Greenberg to Greene is noted by hand on his draft registration card. He began his comic book career in 1941 as an artist for Funnies, Inc. but his career was interrupted by World War II as he enlisted in the service on September 14, 1942. He was discharged on February 1, 1946. One week later, he married Anita Elizabeth Smith, who is thought to have lettered many of Greene's stories in the late 1940s. He worked at Marvel, Holyoke, and Ace before coming to DC Comics in 1955. In addition to this comics work, he is said to have produced the art for a set of anatomy flash cards used by medical and nursing students.
